Its toxicity and danger for human health is still being studied, meanwhile direct influence of environmentally relevant doses was not discovered. These endocrine disruptors are affecting variousreceptorsin bodiesand effecton cardiovascularsystem isnotexcludedeither.The most extensively studied substance is BPA, which is gradually being replaced by its analogs, although these are not extensively researched to the same extent. In this work we focused on influence of bisphenol BP, M and S on differentiation of H9c2 cells, serving as a model with potential negative effects on heart development and its regeneration after damage. Determination of bisphenols in selected beverages by high-performance liquid chromatography

Bisphenols are chemical compounds with a wide range of uses in the chemical industry. They are used in the production of plastic materials as softeners for polymer materials or epoxy resins, as monomers in the synthesis of polycarbonates, or for the elimination of hydrochloric acid in the production of polyvinyl chloride. This thesis deals with the development, validation, and optimization of an analytical method for the determination of selected bisphenols, specifically bisphenol A, bisphenol B, bisphenol F, bisphenol A diglycidyl ether and bisphenol F diglycidyl ether (BPA, BPB, BPF, BADGE, BFDGE), and their metabolites using the high performance liquid chromatography with mass spectrometry, HPLC/MS, and the application of the method to real samples in selected canned beer samples. The following conditions were optimized for chromatographic separation. The Sigma-Aldrich discovery C18 column (150×2,1 mm, 5µm) was chosen. The mobile phase consisted of a mixture of mobile solvent A, which contained 0,05% ammonium hydroxide, and mobile solvent B, which contained of methanol. The flow rate of the mobile phase was 0,240 ml/min. The total analysis time was 22 minutes, using a 5 μl injection of the sample solution with gradient elution. Martin Novák Title of thesis: Development of LC-MS method for determination of selected endocrine disruptors The thesis is focused on the development and optimization of LC-MS method for the determination of twelve endocrine disruptors in amniotic fluid of pregnant women in the third trimestr of pregnancy. There is a suspision that premature births are connected with an increased incidence of endocrine disruptors in amniotic fluid. The analytes were extracted by liquid-liqud extraction with 4-methylpentan-2- on. The analytical method is based on using reverse phase high-performance liquid chromatography with Hypersil C4 GOLD column (2.1 × 100 mm, particles 1.9 µm). For analysis the gradient elution with the mobile phase A (0.5% (v/v) formic acid aqueous solution) and B (0.5% (v/v) formic acid solution in acetonitrile) at a flow rate of 0.5 ml/min, injection 5 µl and column oven temperature 35 řC was used.
